연구분야 소개

(1) 주요 연구분야

본 연구실은 단백질의 구조 분석, 분자동역학, 도킹등을 통해 효소의 안정성, 기질특이성, 활성을 개량하는 연구를 진행한다. 개량된 효소는 유용물질의 생산 및 전환에 이용되며 적용 분야는 매우 다양하다.

▶ 인공금속효소 (artificial Metalloenzyme, Arm) 연구

인공금속효소는 전이금속촉매의 빠른 반응성 및 비자연계 촉매반응과 효소의 선택성 가능 반응을 동시에 구현 한다. 전이금속복합체 탐색, scaffold 단백질 탐색, 금속과 단백질 조합방법 모색, 반응 스크리닝법 구축, 분자동역학을 이용한 구조 분석, 움직임 모식화, 반응 매커니즘 등의 연구를 진행한다. Random mutation 방법을 이용하여 인공금속효소의 라이브러리를 만들고 높은 선택성의 사이클로프로판화 반응을 스크리닝 할 수 있는 효율적인 플랫폼을 만든 연구를 수행하였고 실제 인공금속효소 움직임을 분석하는 연구를 수행중이다.

▶ Molecular modeling을 통한 효소 및 단백질 개량

Molecular modeling (Molecular dynamics, Molecular docking 등)을 통하여 효소 및 단백질의 디자인 전략을 구축 실제 in vitro에 적용하는 연구를 수행한다. 다양한 효소들에 대한 연구를 수행해 왔으며 필요시 Directed evolution, library screening등을 함께 수행한다.
